Ten Beauveria bassiana isolates were characterized using the following parameters: virulence, morphological measurements, germination in solid and liquid media, in vitro growth and physiological changes in liquid media. There were significant differences in mortality of second-instar larvae of the diamondback moth (DBM), Plutella xylostella (Lepidoptera: Plutellidae) and the Colorado potato beetle (CPB), Leptinotarsa decemlineata (Coleoptera, Chrysomelidae) according to isolate used (DBM; 14.51-53.37%, CPB; 25.05-74.07%). The most virulent isolates to DBM and CPB were KCF107 (soil origin) and KCF106 (Chilo suppressalis origin), respectively. Several morphological and physiological characteristics of isolates were evaluated for a possible link to virulence against both insect species. Special mean diameter (2.45-3.58 µm) and area of conidia (5.72-10.25 µm2) were significantly different among isolates. Time for 50% of conidia to germinate (GT50) varied from 11 to 21 h depending on isolate. There was no correlation to virulence of size of conidia, GT50, red pigment production and mycelial growth. Acidity of 7-day-old broth cultures of isolates was positively correlated with virulence. Iranian isolates KCF106 and KCF107 were the most promising and virulent isolates but field testing is needed to further evaluate their potential for use in DBM and CPB management.  相似文献

Abstract

Microbial content formed in bioreactors plays a significant role in the anaerobic process. Therefore, the physicochemical characteristics of microbial content in a modified anaerobic inclining-baffled reactor (MAI-BR) treating recycled paper mill effluent (RPME) were investigated using Fourier transform infrared (FTIR), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), thermogravimetric (TG), and derivative thermogravimetric (DTG) analyses, scanning electron microscopy-energy dispersive X-ray spectroscopy (SEM-EDS), Brunauer-Emmett-Teller (BET), and surface area analyzer. FTIR spectra revealed that the microbial content had stronger characteristic peaks corresponding to alcohols, water, lipids carbohydrates, proteins, and mineral compounds. Calcite, muscovite, and lepidolite were the prevalent mineral phases found by XRD analysis. The elemental of these minerals like C, Ca, N, O, and Si was confirmed by XPS results. The microbial content samples from each compartment showed similar thermal behavior. SEM images showed that straight rod-shaped and Methanosaeta-like microorganisms were predominant, whereas C, O, and Ca were noticed by EDS on the surface of granules. The BET surface areas and pores of granules are found to decline throughout the reactor’s compartment, where Compartment 1 had the largest values. Thus, the findings of this study establish further understanding of the physicochemical properties of microbial content formed in MAI-BR during the RPME treatment.  相似文献

Two glucose-6-phosphate isomerase (GPI) mutants with approximately 60% residual activity in blood compared to wild type have been independently detected in offspring derived from 1-ethyl-1-nitrosourea-treated male mice. Homozygous mutants with about 20% residual activity were recovered in progeny of inter se matings of heterozygotes. However, in both mutant lines the number of homozygous mutants was less than expected suggesting an increased lethality of these animals. Results of linkage studies and of investigations of physicochemical properties of the mutant enzymes indicate point mutations at theGpi-1s structural locus on chromosome 7. Based on these findings the two new alleles were designatedGpi-1s b-m1Neu andGpi-1s b-m2Neu, respectively. The b-m1Neu allele codes for an erythrocyte enzyme which, in the homodimeric form, exhibits a decreased stability toward heat and urea, an altered isoelectric point, normalpH dependence, an increasedK m for fructose-6-phosphate, and increasedK i's for 6-phosphogluconate and 2,3-diphosphoglycerate (2,3-DPG) compared to the wild-type enzyme. The GPI-1sb-m2Neu homodimer, in contrast, is characterized by an even stronger instability, slightly alteredpH dependence, an increasedK i for 2,3-DPG, normal other kinetics, and normal isoelectric point. The different degree of stability of the mutant homodimersin vitro seems to be reflected in a different degree of stabilityin vivo, since GPI deficiency in general is more strongly expressed in the tissues of the homozygousGpi-1s b-m2Neu mutant compared to the homozygousGpi-1s b-m1Neu mutant. The similarity of the mutant enzymes to the allozymes found in human GPI deficiencies indicates the GPI deficient mouse mutants to be excellent models for the human disease.This research was supported in part by Contract BI6-156-D from the Commission of the European Communities.  相似文献

Although avian color preferences have been studied and documented in controlled experiments, they have not been demonstrated under natural conditions in most cases. We hypothesized that avian fruit choice reflects intraspecific variation in fruit characteristics other than color, rather than fruit color differences. By planting one Ilex serrata Thunb. (red form) and one I. serrata forma leucocarpa Beissner (white form), which produce red and white fruits, respectively, at each of five points, we examined the proportion of fruits removed per tree and fruit choice by three avian species based on fruit color and other fruit characteristics. The proportion of fruits removed increased with pulpy sugar concentration and fruit diameter, but it did not differ between fruit colors. The main foragers, resident brown-eared bulbuls Hypsypetes amaurotis, consumed fruits regardless of color, but correspondingly to fruit removal, and appeared to base their fruit choice on pulpy sugar concentration and fruit diameter rather than on color. In contrast, the minor foragers, migrant Daurian redstarts Phoenicurus auroreus (Pallas) and Siberian bluechats Tarsiger cyanurus (Pallas), tended to choose red fruits and were possibly attracted by them. In conclusion, fruit removal per tree reflected individual variation in fruit profitability more strongly than differences in fruit color, even though the individual variation was not remarkable. The importance of color in fruit choice differed based on species, residency status, and major/minor foragers.  相似文献

  • A significant asynchronous phenomenon exists in super hybrid rice because of the differences in spike and spikelet positions, which affect the accumulation and properties of starch. However, little is known about the endosperm enrichment and physicochemical properties of starch in superior and inferior grains in super hybrid rice.
  • Rice YY2640 was selected as study material to investigate the enrichment and physicochemical properties of starch in superior and inferior grains in super rice using semi‐thin sections, X‐ray diffraction and related technologies.
  • Superior grain filling was a continuous process, whereas inferior grain only started 8–10 days after anthesis. The order of starch accumulation starts in the central endosperm, then in the endosperm of the proximal vascular bundle and finally in the aleurone layer. Compared with the inferior grains, the superior grains have a higher 1000‐grain weight, apparent amylose content, total starch content, average starch granule size, relative crystallinity, solubility and a resonance peak ratio at 1022/995 cm?1, whereas the swelling power and ratio of the resonance peak at 1045/1022 cm?1 were lower. The final degree of hydrolysis of HCl, AAG and PPA of the superior grains were significantly lower than those of the inferior grains.
  • The findings indicate that the different physicochemical properties of starch were mainly related to the development order of superior and inferior grains and the spatial enrichment of starch.

The role of the brown-eared bulbul,Hypsypetes amaurotis, as a dispersal agent for seeds of fruiting plants was studied by field observations for two years, in parallel with laboratory experiments on seed germination. Bulbuls consumed fruits of 53 species from 24 plant families. The fruits of these plants had similar color and size, and these characteristics were likely to enhance the feeding efficiency of the frugivore. Laboratory experiments on 20 food plant species demonstrated that: (1) no seeds were injured by passing through the bulbul's gut; (2) seeds that had passed through the bulbul's gut were still able to germinate and (3) fruit pulp reduced germination ability. When pulp was removed by passing through bulbul's gut, or by hand, germination was improved. An estimate of the home range of six bulbuls suggested that they may transport seeds for at least 300 m.  相似文献

Engineering aspects of solid state fermentation have not been available in spite of the recent interest in the technique and the appearance of a number of reviews on this subject. The present state of the art regarding substrate uptake, oxygen transfer, growth characteristics, growth estimation, control systems for maintaining parameters, mathematical models, design of fermenters and automation of fermentations does not provide a detailed insight. The work carried out at the Central Food Technological Research Institute, Mysore, on the development of a large-scale solid state fermenter, comparative cost estimation of SSF and submerged fermentation as well as development of know-how for production of a variety of food enzymes, has led to the commercial exploitation of the technology by industry. Future R&D needs in this area, neglected at present but yet so promising, are indicated.  相似文献
